肿瘤热疗对经尾静脉输注的TIL在裸鼠体内归巢的影响

Effect of Hyperthermia on Homing of Tumor Infiltrating Lymphocytes Transferred via Tail Vein to Tumors in Nude Mice

摘要 目的探讨肿瘤热疗对经尾静脉输注的肿瘤浸润淋巴细胞(TIL)在裸鼠体内归巢的影响。方法裸鼠双后肢大腿荷人颊癌BCaCD885细胞成瘤后,一侧加热,另一侧未加热作为对照,12 h后自尾静脉分别输入经3H-TdR标记的未经处理过的TIL(NSTIL)、经BCaCD885细胞刺激处理过的TIL(NHTIL)及经热处理过的BCaCD885细胞刺激处理的TIL(HSTIL),比较3种TIL在加热与未加热肿瘤中归巢的差异。结果注射后24 h,3种TIL均在经预先加热的肿瘤组织中有更高的聚集,且HSTIL在经预先加热的肿瘤组织中的聚集量远高于另两种TIL,但与肿瘤组织占体重的百分比相比,在肿瘤区定居的TIL数仍属少数。结论加热可增强TIL在肿瘤组织的归巢与定居,此过程可能与TIL对肿瘤细胞的特异性有关。 Objective To investigate the effect of hyperthermia on homing of tumor infiltrating lympbacytes (TILs) transferred via tail vein to tmnors in nude mice. Methods The huecal cancer celt line BCaCD885 xenografts were established in both hind legs of nude mice. One of the legs (tumors) had been treated in 43.5 ℃ water bath for 40 m/nutes, 12 hours before one of three kinds of TILs were transferred via tail vein, which were NH TILs (TILe pre-treated by BCaCD885 cells), HS TILs (TILe pre-treated by BCaCD885 cells and then pre- treated in 43.5℃ water bath for 40 minutes) and NS TILs (TILS without either pretreating). The homing of TILs was determined 24 hours after transfusions. Results More TILs homed to tumors pre-treated by hyperthermia . RS TILs had a higher affinity to the turners as well as a higher tendency to aggregate in tumors than NHTIL and NS TIL. Conclusion Hyperthermia can enhance homing of TILs to tmnor. The potential significance of the results in thermotherapy and immunotherapy for tumors is implied.
