孕激素对MG-63细胞株孕激素受体和c-fos、c-jun的影响

Effects of progesterone on progesterone receptor c-fos and c-jun expression in MG-63 cells

目的观察18-甲左炔诺孕酮对人成骨肉瘤MG-63细胞株增殖、孕激素受体和c-fos、c-jun表达的影响。方法用不同浓度18-甲左炔诺孕酮干预MG-63细胞株,以不含左炔诺孕酮的MEM培养液作对照,MTT法检测细胞增殖,RT-PCR测定孕激素受体(PR)、c-fos、c-jun mRNA表达,Western Blot和免疫组化法测定PR、c-fos、c-jun蛋白质的表达。结果与对照组比较,10-10mol/L、10-8mol/L和10-6mol/L左炔诺孕酮分别增加MG-63细胞增殖达7.72%、24.36%和36.54%(P<0.01或P<0.05)。干预24h后,MG-63细胞中PRA、PRB mRNA和蛋白质表达无明显变化(P>0.05)。干预2h后,左炔诺孕酮增加MG-63细胞中c-fos、c-jun mRNA和蛋白质表达(P<0.01或P<0.05),呈剂量依赖性。结论孕激素可能通过增加c-fos、c-jun的表达来促进MG-63细胞增殖。

[Objective] To investigate the effects of 18-methyl levonorgestrel on proliferation and the expression of progesterone receptor （PR）, c-fos, c-jun in human osteosarcoma cell line MG-63. [Methods] Cell proliferation was tested by MTT assay. The gene expression of PR, c-los and c-jun was measured by semi-quantitive reverse transcription-polymerase chain reaction （RT-PCR）. The protein expression of PR, c-fos and c-jun was measured by Western blot or immunocytochemistry. [Results] 10^-10 mol/L, 10^-8 mol/L, 10^-6 mol/L levonorgestrel increased the cell proliferation by 7.72%, 24.36%, 36.54% respectively （P 〈0.01 or P 〈0.05）. Levonorgestrel had no effect on PRA and PRB mRNA and protein expression after 24 h of treatment. While the c-fos and c-jun mRNA and protein were upregulated in a dose-dependent manner after 2 h of treatment （P 〈0.01 or P 〈0.05）. [Conclusions] Levonorgestrel promotes the expression of c-fos and c-jun, and results in MG-63 cells proliferation.
