用PCR-SSCP方法检测间皮瘤组织中p53基因点突变

Detection of point mutations of p53 gene by non isotopic PCR SSCP in paraffin embedded malignant mesothelioma tissue

摘要 为研究石棉纤维在人恶性间皮瘤病因学中的作用,检查了青石棉污染区12例间皮瘤DNA样品的p53基因点突变。应用非同位素PCR-SSCP方法,聚丙烯酰胺凝胶电泳分析了p53基因5,6,7,8外显子的4个DNA片段,结果显示:4例有异常移动的单链DNA电泳带,其中2例位于外显子5,2例位于外显子6。 To investigate the role of asbestos fibers in the etiology of human malignant mesothelioma,the point mutation in p53 gene DNA sample of 12 patients from crocidolite contamination area was examined.Non isotopic PCR SSCP was used to detect point mutation of p53 exons 5,6,7,8 of p53 gene in paraffin embedded mesothelioma tissues.After 4 DNA fragments of p53 exons 5 ̄8 in p53 gene were put to undergo electrophoresis in a nondenaturing polyacrylamide gel,the abnormal migration of the single stranded DNA was identified in 4 out of 12 cases.The mutations of 2 cases were found in exon 5,2 cases in exon 6.The result suggested that point mutation of p53 gene may be present in human malignant mesothelioma.
