摘要 目的:建立牛主动脉内皮细胞(BAEC)的体外研究模型,为体外研究内皮细胞的病理和生理过程提供重要手段。方法:用酶消化法分离BAEC,在pH为7.2的DMEM培养基中培养,鉴定内皮细胞中特异表达的第Ⅷ因子;以计数法测细胞的增殖能力,同时测定细胞的迁移。结果:细胞呈三角形,单层生长,第Ⅷ因子表达阳性,增殖呈指数生长,迁移正常。结论:提示牛主动脉内皮细胞的体外模型建立成功。 Objective:To establish isolation model of bovine arterial endothelial cells in vitro for further pathological and physiological studies. Methods: Endothelial cells from bovine thoracic aorta were isolated using enzyme digesting method, identified with Factor Ⅷ. Cell proliferation and migration characterization were also investigated. Results: The isolated cells showed characteristic triangle in shape, positive Factor Ⅷ staining and with normal migration ability. Conclusion: The isolation model of bovine arterial endothelial cells is successful.
