摘要
目的:观察帕金森模型鼠中脑D-谷氨酸的变化情况,探讨兴奋性神经递质右旋对映体对谷氨酸能神经元的损伤作用以及与帕金森发展过程的相关性。方法:成年C_(57)BL雄性小鼠随机分为对照组、模型组。模型组腹腔注射1-甲基-4-苯基-1.2.3.6-四氢吡啶(1-methyl-4-phenyl-1.2.3.6-tetrahydrophridine,MPTP)制备帕金森病模型鼠。采用柱前衍生高效液相色谱方法检测中脑D-谷氨酸含量,采用免疫组织化学ABC法观察黑质和尾壳核谷氨酸能神经元的变化。结果:与对照组相比,模型组动物注射MPTP 1d后中脑组织中D-谷氨酸含量明显升高,3 d后逐渐降低;3 d后谷氨酸能神经元数量明显减少。结论:中脑组织中D-谷氨酸可能具备与L型对映体相同的兴奋毒性,并参与帕金森的发病机制。
Objective: To explore the toxicity of the D-glutamate (D-Glu) on glutamatergic neurons and its correlation to the progress of Parkinson's disease (PD) through the change of D-Glu in the midbrain of mice with PD. Methods: Adult C57BL male mice were randomly divided into the control group and PD model group. The PD model was established by intraperitoneal injections with MPTP. The change of D-Glu in midbrain was detected by using highperformance liquid chromatography method. The Glu-neurons in the Substantia Nigra and Nu. of caudate-putamen were observed by using immunohistochemical method. Results: After injection with the MPTP, Glu-neurons were reduced gradually, but D-Glu in the midbrain was increased significantly compared with that in the control group. Conclusion: D-Glu might have the same toxicity on Glu-neurons as L-Glu in the progress of PD.
