摘要
目的对临床分离鲍曼不动杆菌进行耐药基因克隆。方法利用已知Acinetobacter sp.ADP1全序列设计引物,通过PCR扩增,进行耐药基因的克隆;对重组菌株进行药敏分析。结果克隆出一个耐药基因,该基因编码的蛋白对氯霉素、四环素和链霉素等抗生素及溴化乙锭、罗丹明6G、脱氧胆酸钠和十二烷基硫酸钠等抗菌剂具有很强耐药性。结论克隆到的耐药基因为一多重耐药基因。
Objective To clone a drug - resistant gene of Acinetobacter baumanni isolated from 35 clinical patients. Methods The primers were designed based on genomic DNA of Acinetobacter ADP1, and PCR amplification of drug - resistant gene was carried out using genomic DNA ofAcinetobacter baumannii 35; The minimal inhibitory concentrations (MICs) of various drugs were determined by serial two- fold dilution method. Results A drug- resistant gene was cloned and the transformant harboring recombinant plasmid showed high resistance to chloramphenicol, tetracycline, streptomycin, ethidium bromide, rhodamine 6G, DC and SDS. Conclusion The cloned gene is a multiple resistance gene.
