丙酮法和碱法脱脂对胶原蛋白生产的影响 被引量:10

摘要 文章比较了碱法和丙酮法对骨粉的脱脂效果,测定了两种方法生产骨胶原蛋白的量,并对提取的骨胶原蛋白进行了SDS-PAGE电泳分析。结果表明,在4℃下,NaOH溶液在1.0 mol/L作用3 d时,脱脂率最高达96.83%,丙酮脱脂作用3 d时的脱脂率为98.85%;本试验中提取的胶原蛋白是典型的Ⅰ型胶原蛋白,其纯度为74.33%。两种方法比较,丙酮法脱脂效果好,所得样品强度高,且成本低。 A comparison of the effect of degreasing from bone with alkali and acetone was made and tide collagen production was measurated by the two methods. Finally the SDS-PAGE was used to analyze the purity of the collagen extracted. The results showed that the degreasing rate was the highest of 96.83% after dipping the bone powder in 1.0 mol/L NaOH solution for 3 days at 4℃, and the degreasing rate was the highest of 98.85% after dipping in the acetone for 3 days. The extracting collagen was the typical type Ⅰ collagen and its purity was 74.33%. Compared with alkali method,acetone method got better degreasing effect and collagen purity,and was more economic.
