MBP诱导小鼠免疫细胞凋亡的体内研究

PRELIMINERY EXPLORATION OF APOPTOSIS IN MURINE IMMUNOCYTES INDUCED BY MBP IN VIVO

为初步探讨ＭＢＰ与免疫细胞凋亡的关系，实验设置了超抗原金黄色葡萄球菌肠毒素Ｂ（ＳＥＢ）作为阳性对照，普通蛋白抗原ＢＳＡ为阴性对照。实验方法以检测ＤＮＡ断裂为主，配以形态学观察。结果表明，ＭＢＰ处理组免疫细胞ＤＮＡ经电泳可见“典型”的梯状带；通过比色法和流式细胞术定量检测了ＤＮＡ断裂的数值。结果显示，ＭＢＰ处理组ＤＮＡ断裂百分数与ＳＥＢ组无明显区别（Ｐ＜０．０５）。同时形态学观察表明ＭＢＰ处理组凋亡细胞数目与ＳＥＢ组基本相同。上述结果表明ＭＢＰ具有类似超抗原ＳＥＢ的效应。

Investigate whether the MBP play a role as superantigen to induce apoptosis.Mice are divided into three groups.One is MBP,the two others,SEB and BSA,are used as positive and negative control,respectively.DNA fragmentation is demonstrated quantitatively by agarose gel electrophoresis,colorimetry and flow cytometry,accompanying the qualitative morphological observation.The results show that the DNA of cells in MBP group displays a charactersitic DNA fragmentation.The percentage of DNA fragmentation in MBP group is similar to that of SEB group.The morphological observation is consistent with the above DNA analysis.As mentioned above,these results could provide a novel information that MBP has a similar effect as superantigen SEB.
