摘要
目的为了研究透明颤菌在微生物发酵工业生产中的应用,对东北寒带地区透明颤菌的理化特性进行研究。方法透明颤菌的分离采用培养基(PYA)富集培养与形态观察的方法,采用分光光度计、光扫描与CO差示光谱确定透明颤菌的生理生化性质。结果研究分离的透明颤菌在蛋白胨0.4%,酵母0.6%,NaAc 0.02%,pH7.8,34℃,150 r/min时可获得较大生长速率,NaAc浓度为1%,pH7.5-8.0时菌体生长最快。透明颤菌在稳定期血红素含量约为每克湿菌中11 nmol,样品在5000 r/min离心后出现血红蛋白的吸收峰。结论分离培养出寒带地区生长的透明颤菌,并确定了其理化性质,为在发酵工业生产中应用透明颤菌奠定了参考依据。
Objective In order to study using of Vitreoscilla in microbiological fermentation industry, the physiological and chemical characteristics of Vitreoscilla were studied from eastnorth cold region. Methods Screening of Vitreoscilla by using culture (PYA) and pattern observe. The physiological and chemical characteristics of Vitreoscilla were determined by optical density (A) ,CO difference spectra and the pyridylheme spectra assay. Results The optimum growth conditions for Vitreoscilla were in medium of tryptone 0.4% , yeast 0.7% , NaAc 0.02% , pH 7.8 - 8.0 and 34 ℃ in a shaker incubator set at 150 r/min. VitreosciUa heme content was about 11 nmol/g in humid germ,sample hemoglobin absorbance peak was discovered in after 5000 r/min centrifugation. Conclusions This treatise had separated Vitreoscilla Eastnorth strain. Its physiological and chemical characteristics were identified, this study has provided a reference basis for Vitreoscilla Hemoglobin using at fermentation industry.
