沙蚕毒素类农药巴丹对Vero细胞的毒性及活性氧在细胞损伤中的作用 被引量:3

摘要 目的研究沙蚕毒素类农药巴丹对Vero细胞的毒性作用及活性氧(ROS)在其引起的细胞损伤中可能发挥的作用。方法以10μmol/L~1000μmol/L浓度的巴丹作用于Vero细胞4h、8h、12h、24h、48h后,通过MTT法观察细胞存活率,并在倒置显微镜下观察对细胞形态的影响。不同浓度作用8h、16h、24h、48h后通过乳酸脱氢酶(LDH)漏出率检测法观察巴丹对Vero细胞的杀伤率。不同浓度作用1h后,荧光分光光度计检测巴丹对Vero细胞内ROS含量的影响。结果能引起Vero细胞LDH漏出率升高、细胞增殖率下降、细胞内ROS水平升高,呈时间、剂量依赖关系。结论巴丹对Vero细胞存在明显的毒性作用,ROS在其引起的细胞损伤可能发挥重要作用。 Objective To investigate the cytotoxicity of cartap and the role of reactive oxygen species (ROS) on its toxic effects in Vero cell line. Methods Vero cells were cultured in presence of cartap at the concentration of 10μmoi/L-1000μmoi/L for 4h, 8h, 12h, 24h, 48h and then the MTT method was used to evaluate the cell viability. At the same time the morphological changes of cultured cells were observed. Lactate dehydrogenase (LDH) leakage was measured by colorimetric method to estimate the cell mortality caused by the toxicity of cartap. The fluorescent intensity of ROS was determined by spectrofluorometer after Vero cells treated with cartap at different concentrations for I h. Results Cartap could obviously decrease the viability rate and increase the mortality of Vero cell in dose-dependent and time-dependent manner. In the meantime, ROS level in the cell was increased significantly in dose-dependent and time-dependent manner as well. Conclusion Cartap has a significant cytotoxicity effect on Veto cell, and the cytotoxic mechanism may be that it could induce cellular injury by increasing ROS level.
