摘要 目的:探讨大鼠重症急性胰腺炎模型制备方法的改进。方法:36只健康SD大鼠随机分为模型组(n=18)和对照组(n=18)。应用硬膜外导管经十二指肠乳头逆行胰胆管穿刺注射5%牛磺胆酸钠诱导大鼠重症急性胰腺炎;使用创面封闭胶封闭穿刺孔。两组大鼠分别于术后3、6、12h检测腹水量、血清淀粉酶活性,观察胰腺大体及光镜下病理改变并评分。结果:模型组胰腺呈出血坏死性改变,3、6、12h的腹水量,血清淀粉酶活性,大体及镜下评分均显著高于对照组(P<0.05)。结论:联合应用硬膜外导管和创面封闭胶对逆行胰胆管穿刺注射法加以改进,具有创伤小,操作简单,诱导成功率高,模型稳定性好等优点,是一种较理想的制模方法。 Objective:To study related technique improvement on animal model with severe acute pancreatitis in rats. Methods:The Sprague-Dawley rats were randomly divided into model or control group. Severe acute pancreatitis animal model was induced by injection of 5% sodium deoxycholate through retrogradely common biliopancreatic ducts via papilla duodeni with epidural catheter, puncture hole was closed by medical sealant glue. Ascites and blood sample was obtained at 3、6 and 12 hours respectively. General and histological changes of pancreas was evaluated by the corresponding score system. Results:Pancreatic changes in the model group was hemorrhagic and necrotizing. The ascites volume, serum amylase activity, general and histological scores in model group was significantly higher than that in control group (P〈0.05). Conclusion:Combined epidural catheter and medical sealant glue in the experiment could minimize the trauma and simplize the procedures.It might be a high success rate, reliable model and it is an ideal severe acute pancreatitis animal model to be recommended.
