RP-HPLC梯度洗脱测定至圣保元丸中升麻素苷和5-O-甲基维斯阿米醇苷的含量

目的测定至圣保元丸中升麻素苷和5-O-甲基维斯阿米醇苷含量。方法样品用甲醇提取,采用反相高效液相色谱法,色谱条件:Luna C18柱(250nm×4．60mm,5μm)为分析柱,甲醇-乙腈-0．04％醋酸铵为流动相进行梯度洗脱;检测波长254nm,流速1.0ml·min-1;对4批样品进行了主成分升麻素苷和5-O-甲基维斯阿米醇苷的总量测定。结果每克至圣保元丸中升麻素苷、5-O-甲基维斯阿米醇苷的含量分别为0．159mg,0．11mg。回收率分别为104．16％,95．60％。结论本方法测定至圣保元丸中的升麻素苷、5-O-甲基维斯阿米醇苷的含量准确、快速。

OBJECTIVE To determine the content of Prim- O-Glucnylcimifugin and 5-O-Methylvismmiside in Zhishengbaoyuan Pill. METHODS RP-HPLC method was usded by Kromasil C18（250nm × 4.60mm, 5μm）in methanol-acentonitrile-0. 04% Acetate ammomum as a mobile Phase （1.0ml · min^-1 ）. The UV detection wavelength was set at 254nm. 4 lot of samples were detecded. RESULTS The content of Prim-O- Gluconylcimifugin and 5-O-Methylvismmioside were 0. 159 and 0.11mg in 1g Zhishengbaoyuan Pill. The recoverys were 104. 16% and 95.60% ,respectively. CONCLUSION The method was simple,rapid accurate and reproducible.
