摘要
对离子束诱变筛选得到的阿魏菇菌株,进行了胞内多糖提取纯化试验,分别对影响多糖提纯的提取条件,醇析条件,盐析条件,去蛋白效率,H2O2脱色进行了研究,并利用回归模型对工艺条件的最优组合、单因子效应进行了探讨。结果表明,胞内多糖最佳提纯工艺为,浸提料液比15,温度100℃,3h,提取3次,置-20℃冰冻后可初步去除不溶杂质,然后向多糖溶液中加入试剂硫酸铵和无水乙醇析出多糖(加入条件为:硫酸铵和多糖液m/m1:5;无水乙醇和多糖液v/v1:1),静置8h,H2O2脱色6h。
This research showed a high yield Pleurotus ferulae Lenzi strain by low energy ion implantation that extraction methods of polysaccharide from mycelium were studied by orthogonal experiments, rotation and combinational design of quadric regression the basis of determination of the extraction rates of Folysaccharides.The extracting conditions, ethanol precipitation, saline concentration, remove protein,H2O2 discolor had been discussed, The results showed that the optimal condition are 15 times water ,100 ℃,3 h,extraction of polysaccharides 3 times, precipitate condition is Ammonium sulfate with ethanol(Ammonium sulfate and polysaccharide solution is 1:5(m/m), ethanol and polysaccharide solution is 1:1 (v/v))8 h, H2O2 discolor 6 h.
