摘要
利用自制高压静电场装置,对啤酒酵母在芒刺高压静电场下生活微环境的变化进行了测定,结果表明,啤酒酵母经芒刺高压静电场处理后,菌液电导率、pH值随处理参数的改变发生规律性的变化;啤酒酵母的外观形态遭到破坏,中部出现凹陷,表面出现褶皱;经电场处理并去除菌体后的上清液中的蛋白质和核酸含量与未经处理的对照上清液相比有显著增加。可以认为,细胞膜在高压静电场下受到破坏,细胞质出现渗漏是啤酒酵母致死的主要原因。
In this study, the changes of living circumstance of S.cerevisiae is treated by needle and plate high-voltage electrostatic field (HVEF) made by myself, the results showed that, conduction rate and pH value changed regularly after S.cerevisiae treated by HVEF, the microscope videotape showed configuration of S.cerevisiae are terribly destroyed by HVEF. The results of the detection of cellular protein and nucleic acid outflowing variety showed needle and plate HVEF has a destructive effect on cell. It can be said that the HVEF to destroyed the membrane which led to cytoplasm leakage is the main cause of death of S.cerevisiae.
