摘要
研究了一种快速检测蔬菜中农药残留(有机磷)的方法。根据酶抑制法的原理,对反应温度、pH、反应时间、显色剂及样品提取液等条件进行优化,建立了以聚苯乙烯反应板为反应载体的农药(有机磷)快速检测方法,其最小检出量为0.245μg。
A method of rapid detecting organophosphorus residues in vegetables wasstudied. The condition of temperature, pH value, reaction times, colored and sample extractions in reaction were optimized according to the theory of enzyme inhibition. The results show that the plant-esterase is diluted with 0.1 mol/L phosphate buffer contained 0.1% bovine serum albumin and glutaraldehyde(1: 1,v/v),Which immobilized at 37 ℃ for 24 h.The sample is extracted with 0.1 mol/L(pH=6.2)phosphate buffer, colored by o-dianisidine in 0.1 mol/L (pH=6.6) phosphate buffer and terminated with 10 % trichloroacetic acid methanol solvent, detected at 570 nm.The recovery rate is 87 % to 10 % and minimum limited of the assay is 0.245 μg.The method was established to rapid determine the pesticide residues in vegetables base on polystyrene plate as a reaction carrier.
