摘要
目的探讨葛根素(puerarin)对溶血磷脂酰胆碱(lysophosphatidylcholine,LPC)损伤家兔血管内皮依赖性舒张功能的影响。方法采用离体血管环张力实验法检测乙酰胆碱诱导的内皮依赖性舒张反应来评价LPC对血管内皮功能的损害和葛根素的保护作用。结果分别用2.5-10 mg/L LPC孵育血管环30 min,出现剂量依赖性抑制乙酰胆碱诱导的内皮依赖性舒张反应。用0.25-1 g/L葛根素分别孵育血管环15 min,再与5 mg/L LPC共同孵育30 min,明显改善LPC所致的血管舒张功能的损害。结论葛根素对LPC所致的血管内皮依赖性舒张功能的损伤具有明显的保护作用。
Objective To investigate effects of puerarin on impaired endothelium- dependent relaxation induced by lysophosphatidylcholine (LPC) in rat thoracic aorta. Methods The endothelium- dependent relaxation response to acetylcholine of aortic rings were measured by recording isometric tension after the rings were exposed to LPC in the absence or presence of puerarin to estimate injury effect of LPC and the protective effect of puerarin on aortic endothelium, respectively. Results Exposure of aortic rings to LPC (2.5 - 10 mg/L) for 30 min induced a significant concentration - dependent inhibition of endothelium - dependent relaxation to acetylcholine. Pre - incubation of aortic rings with puerarin (0.25 - 1 g/L) for 15 min and then co - incubation of the rings with LPC (5 mg/L) for another 30 rain significantly attenuated the inhibition of endothelium- dependent relaxation induced by LPC. Conclusion Puerarin can protect the impairment of endothelium - dependent relaxation induced by LPC in rabbit aorta.
