摘要
[目的]观察β肾上腺素受体激动剂异丙肾上腺素对心房钠尿肽(ANP)分泌的影响,探讨其机制.[方法]采用搏动的心房灌流模型及放射免疫技术观察异丙肾上腺素对ANP分泌、环磷酸腺苷(cAMP)逸出量及心房动力学变化的影响.[结果]异丙肾上腺素可明显抑制ANP的分泌,显著增加心房搏出量及环磷酸腺苷逸出量;蛋白激酶非选择性抑制剂Staurosporine可减缓异丙肾上腺素对ANP分泌的抑制效应.[结论]β肾上腺素受体激动剂异丙肾上腺素抑制ANP分泌作用由cAMP依赖性信号传导途径来实现.
OBJECTIVE To investigate the effects of β-adrenoceptor activator, isoproterenol, on atrial natriuretic peptide (ANP) secretion and it's machanism. METHODS The experiments had done in isolated perfused beating rabbit atria. ANP and cAMP were measured by radioimmunoassay in the atrial perfusate in real-time base. RESULTS Isoproterenol could inhibite ANP secretion and increase atrial stroke volume and cAMP efflux. Staurosporine, a non-selective protein kinases inhibitor, could attenuate isoproterenol-induced inhibition of atrial ANP secretion. CONCLUSION Isoproterenol inhibits atrial ANP secretion via the cAMP-dependent signaling pathway.
