延边地区朝鲜族和汉族PPAR-γ 2基因Pro 12 Ala多态性与2型糖尿病关系的研究 被引量:11

摘要 [目的]探讨延边地区朝鲜族和汉族人群中PPAR-γ 2外显子B的Pro 12 Ala多态性与2型糖尿病的相关性.[方法]应用聚合酶链反应-限制性片段长度多态性技术(PCR-RFLP),对183例2型糖尿病患者和145例正常人群PPAR-γ 2基因外显子的Pro 12 Ala多态性基因型进行对照分析.[结果]朝鲜族和汉族正常人群的PPAR-γ2基因Pro 12 Ala多态性的A等位基因频率分别为4.1%,3.5%,两民族间无显著性差异;2型糖尿病组与糖耐量正常组的A等位基因频率分别为3.7%,3.9%,两组间无显著性差异.[结论]PPAR-γ 2基因外显子的Pro 12 Ala多态性在朝鲜族及汉族之间无差异;PPAR-γ2基因Pro12Ala变异与2型糖尿病无明显相关性. OBJECTIVE To determine frequencies distribution of peroxisome proliferator activated receptor γ2 (PPAR-γ2 ) gene Pro 12 Ala polymorphism genotype and allele in Korean-Chinese nationality and Han-Chinese nationality in Yanbian area, and relationship with type 2 diabetes mellitus. METHODS Polymerase chain reaction-restriction fragment length polymorphism was used to test a coding variant Pro 12 Ala of PPAR-γ2 gene in 145 cases of Han-Chinese nationality and Korean-Chinese nationality with normal glucosetolerant (NGT), and 183 cases of Han-Chinese nationality patients and Korean-Chinese nationality patients with type 2 diabetes mellitus (DM), Case-contrast analysis was used to study the association between Pro 12 Ala polymorphism andtype 2 DM. RESULTS The Aallele frequencies of NGT subjects were 4.1% for Korean-Chinese nationality, and 3.5 % for Han-Chinese nationality, and no significant difference was found between two groups. The Aallele frequencies of type 2 DM patients were 3.7% for Korean-Chinese nationality, and 3.9% for Han-Chinese nationality, and no significant difference was found between two groups too. CONCLUSION There was no difference in frequencies distribution of PPAR gene Pro 12 Ala polymorphism between Han-Chinese nationality and Korean-Chinese nationality in Yanbian area, and the Pro 12 Ala polymorphism was not associated with type 2 DM.
