高效液相色谱法测定熊胆粉中牛磺熊去氧胆酸含量

Assaying of tauroursodeoxycholic acid in bear biliary drainage powder by HPLC

[目的]建立熊胆粉中牛磺熊去氧胆酸的含量测定方法．[方法]采用高效液相色谱法测定熊胆粉中牛磺熊去氧胆酸的含量，色谱柱为Allsphere-ODS-Ct8柱（4．6mm×250．0mm，5μm），流动相为甲醇-30mmol/L磷酸二氢钠溶液（65：35，pH值为3．0），检测波长为205nm．[结果]样品中各成分分离良好，进样量在10～35μg范围内呈较好的线性关系（r=0．9996）．[结论]利用高效液相色谱法测定熊胆粉中牛磺熊去氧胆酸的含量具有专属性强及结果准确等特点，可作为熊胆粉的质量控制方法．

OBJECTIVE To establish the method of determination of tauroursodeoxycholie acid in bear biliary drainage powder. METHODS The content of tauroursodeoxyeholie acid in bear biliary drainage powder was determined by HPLC. Chromatorgraphy was performed on a Allsphere-ODS-C18 column, and the mobile phase was methanol-30 mmol/L NaH2PO4（65：35, pH 3.0） ,and the wavelength of detection was 205 nm. RESULTS Different components of specimen were separated favorably, and HPLC exhibited a fine linear response of tauroursodeoxycholic acid ranged from 10- 35μg（ r = 0.999 6）. CONCLUSION HPLC is precision and reproducibility, and that can be used as a specialized determination of tauroursodeoxycholic acid in drainage powder.
