摘要
目的探讨β-连环蛋白(β-cat)在卵巢癌组织中的表达及意义。方法①应用4097点基因芯片对5例卵巢癌及5例正常卵巢组织进行基因表达谱分析,筛查差异表达基因。②对60例卵巢癌组织、24例卵巢瘤组织及24例正常卵巢组织石蜡标本进行免疫组织化学方法染色,检测β-cat的表达情况,分析β-cat的表达与临床分期及组织分级的相关性。结果①基因芯片筛查出卵巢癌差异表达基因58个,其中基因表达增高(上调趋势)30个,基因表达降低(下调趋势)28个,β-cat为表达增高基因。②β-cat在卵巢癌组织中的阳性表达率为88.33%,在卵巢瘤组织中的阳性表达率为33.33%,两组比较有显著性差异(P<0.05);β-cat在正常卵巢组织中的阳性表达率为20.83%,两组比较有显著性差异(P<0.05);临床分期越晚,β-cat阳性表达率越高,卵巢癌Ⅳ期β-cat阳性表达率为93.87%,卵巢癌Ⅰ期β-cat阳性表达率为77.77%,两组比较有显著性差异(P<0.05);不同组织分级β-cat阳性表达率不同,比较无显著性差异(P>0.05)。结论β-cat在卵巢癌组织中表达增高,β-cat表达程度与临床分期正相关,与病理分级无相关性。
Objective To study the expression of β-catenin(β-cat) and their significance in ovarian cancer. Methods ①We used cDNA microarray which contain 4097 genes to analyze gene expression pattern in tissue samples from 5 cases human ovarian cancer to screen genes of differential expression. ②Immunohistochemistry was used to detect the expression of β-cat in 60 cases of ovarian cancer and 24 cases of ovarioncus and 24 cases of normal ovarian tissue ,to analyze the dependability among the expression of (β-cat) and clinical stage and cell differentiatiun.Results ①58 Genes of differential expression were detected in ovarian cancer, expression of 30 genes increased (upregulated), expression of 28 genes decreased (down- regulated), β-cat is upregulated gene. ②The positive rates of β-cat in ovarian cancer and ovarioncus was 88.33% and 33.33% ,there were significant difference between two groups(P 〈 0.05) .The positive rates of β-cat innormal ovarian tissue was 20.83% ,there were significant difference between two groups( P 〈0. 05 ). The positive rates of β-cat was upregulated along with the progressing of clinical stage( P 〈 0.05). The positive rates of β-cat in ovarian cancer stage Ⅳ and stage Ⅰ was 93.87% and 77.77% ,there were significant difference between two groups( P 〈 0.05) ,but no differences were found in relation to cell differentiation ( P 〉 0.05). Conclusion The expression of β-cat was significantly increased in ovarian cancer,there was positive correlation between the expression rate of β-cat and clinical stage,but there wasn't dependability with cell differentiation.
