摘要
目的 观察侧脑室给予noggin对海人酸(kainie acid,KA)诱导的颞叶癫痫大鼠空间学习记忆的影响。方法 侧脑室注射KA诱导大鼠癫痫持续状态(status epileptieus,SE)发作后,连续7d侧脑室注射noggin,2月后大鼠自发性癫痫出现时,Morris水迷宫试验进行行为学检测,尼氏染色检测海马神经元丢失。结果KA诱导大鼠颞叶癫痫成功后,海马CA,、CA。区神经元丢失明显,Morris水迷宫实验中大鼠的逃逸潜伏期延长[(27.38±4.75)S],穿过原平台位置次数[(5.35±1.45)次]和在原平台象限探索时间百分率[(28.73±3.10)%]下降。侧脑室注射noggin的大鼠,海马神经元丢失减少,Morris水迷宫试验中大鼠的逃逸潜伏期缩短[(18.45±2.32)S],穿过原平台位置次数[(9.80±1.53)次]和在原平台象限探索时间百分率[(46.58±3.70)%]增加。结论 KA致颞叶癫痫大鼠侧脑室给予noggin可改善大鼠的空间学习记忆能力,可能与noggin能保护海马神经元有关。
Objective To study the effect of noggin on learning and memory of rats with temporal lobe epilepsy induced by kainic acid. Methods Model of temporal lobe epilepsy(TLE) in rats was established by intracerebroventricular injection of KA. Noggin was injection into lateral cerebral ventricle for 7 days after status epilepticus(SE) was induced. The rats developed spontaneous recurrent seizures (SRSs) 2 months after SE, thereafter morris water maze test was performed and nissl staining was observed in CA3 , CA4 region. Results The model of TEL in rats was established successfully 2 months after SE, and all the rats developed SRSs. The neuronal cell loss mainly occurred in CA3, CA4 region. In the Morris water maze test, latency to find a hidden platform(27.38 ± 4.75) s was longer than NS control group, the times of crossing the situation of platform(5.35 ± 1.45) and the time percent of swimming in northeast( 28.73 ±3.10)% obviously decreased. With intracerebroventricular injection of noggin,escape latency( 18.45 ± 2.32) s was shorter than KA control group, the neuronal cell loss in CA3 , CA4 region obviously decreased,and the times of crossing the situation of platform(9.80 ± 1.53 ) and the time percent of swimming in northeast(46.58 ± 3.70) % obviously increased. Conclusion Intracerebroventricular injection of noggin may decrease learning and memory disturbance in rats with TLE induced by KA. The effect may be due to the protection of noggin on neuronal cell in CA3 , CA4 region.
