电磁辐射致海马损伤的比较蛋白质组学研究 被引量:9

摘要 目的研究三种不同频率电磁波辐照后海马组织蛋白质表达谱的差异。方法采用双向凝胶电泳(2-DE)—质谱鉴定—生物信息学分析的比较蛋白质组学方法研究辐照后6 h海马组织总蛋白的差异,通过W estern blot和RT-PCR技术分别在蛋白和基因水平对差异蛋白质进一步检测验证。结果三种电磁辐射致海马蛋白质表达谱的变化基本相似。经图像分析共筛选到45个差异蛋白点,其中26个差异蛋白点得到了初步鉴定。对于信号转导相关差异蛋白raf激酶抑制蛋白(RKIP),W estern blot和RT-PCR进一步验证了2-DE结果。辐照后6 h RKIP蛋白及其mRNA表达均明显下调,各照射组未见明显差异。结论三种电磁辐射对海马的致伤机制具有相似性,其损伤程度呈频率依赖性。RKIP可能在辐射致海马损伤中发挥重要作用。 Aim To investigate the difference of hippocampus proteome maps after radiation by three electromagnetic waves of different frequency. Methods Comparative proteomic approach based on two di- mensional gel electrophoresis (2-DE) , mass-spectrum identification and bioinformatics analysis was used to study the difference of hippocampus total protein at 6 h after electromagnetic radiation exposure. Western blot and RT-PCR technique was applied to detect the differential protein for further verification on the level of protein and gene respectively. Results The three kind of electromagnetic radiation changed the hippocampus protcome map similarly. 45 differential protein spots were found by image analysis, among of which 26 spots were identified initially. For raf kinase inhibitor protein ( RKIP), which was one of the differential proteins and correlated with signal transduction, Western blot and RT-PCR results further verificated 2-DE results, The expression of RKIP protein and mRNA was down regulated significantly at 6h after radiation, and no significant difference was observed between any radiation groups. Conclusion The injury mechanisms of hippocampus by the three kind of electromagnetic radiation possessed similarity, and the degree of injury showed frequency dependence. RKIP may play an important role in the hippocampus injury by radiation.
