摘要
目的探讨中草药白花蛇舌草(HDW)醇提取物在体外对人白血病细胞株的增殖抑制和凋亡作用。方法以白血病U937细胞为研究对象,以不同浓度和不同时间HDW醇提取物为干预因素,用MTT法检测HDW醇提取物对U937细胞的抑制作用;用细胞形态学、流式细胞术、DNA电泳观察细胞凋亡情况。结果U937细胞经HDW醇提取物作用后,呈时间和剂量依懒性。光镜观察可见细胞膜完整、核碎裂、凋亡小体等形态学特征,DNA电泳呈现典型的梯形条带,流式细胞仪分析表明0.5,1,2,4 mg/mL的HDW醇提取物使U937细胞发生凋亡率分别为4.18%,56.96%,82.43%,56.41%。结论HDW醇提取物抗肿瘤效应强,同时抑制U937细胞的增殖及诱导细胞凋亡。
OBJECTIVE To investigate the effect of ethanol extract of Oldenlandia diffusa Wiild on the proliferation and apoptosis of U937 cells. METHODS By MTT colometric assay, the inhibitory effect of ethanol extract of Oldenlandia diffusa on U937 cells was tested. Expesured log phase growth U937 cells to different concentratioias of ethanol extract of Oldenlandia diffusa for different time, respectively ,and then test the inhibitory effect by modified tertrozalium salt(MTT) assy. The apoptosis of Oldenlandia diffusa induced on U937 cells was detected by optics microscope, flow cytometry and gel electrophoresis of DNA. RESULTS The antiproliferation effects were observed following exposure of ethanol extract of Oldenlandia diffusa. It showed a very substantial differences in the dose and duration dependency of the observed antiproliferative and cytotoxic effects. The U937 cells treated by ethanol extract of Oldenlandia diffusa are observed apeptosis characteristic morphological changes such as complete cell membrane, nuclear fragmention apeposic bodies and found trapezoidal belt with DNA. When ethanol extract of Oldenlandia diffusa treated U937 cells at the dose of 0.5,1,2, 4 mg/mL for 48 hours ,the numbers of the apoptotic cells were respectively 4.18% ,56.96% ,82.43%, 56.41%. CONCLUSION Ethanol extract of Oldenlandia diffusa has the ability to inhibit the proliferation and induce the apoptosis of U937 ceils.
