益肾活血方剂对糖尿病大鼠肾组织胰腺组织氧化应激抑制作用的实验研究

摘要 目的:探讨益肾活血方剂对糖尿病大鼠肾组织、胰腺组织氧化应激的抑制作用。方法:将SD大鼠分为4组:正常对照组(A组)、低剂量组(B组)、高剂量组(C组)、糖尿病组(D组)。B、C、D3组制成糖尿病模型,B组予以中药益肾活血方剂18g/kg,C组予以26g/kg。用药6周后,断头处死动物,立即取左肾和胰腺用生理盐水制成10%的组织匀浆,测定肾组织、胰腺组织丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)活力和超氧化物歧化酶(SOD)的活力。结果:糖尿病大鼠MDA含量明显增高,GSH-Px、SOD活力明显降低,此益肾活血方剂能够增高肾组织GSH-Px、SOD活力和胰腺组织的GSH-Px活力。结论:此方剂通过增高肾组织GSH-Px、SOD活力和胰腺组织的GSH-Px活力起到抗氧化的作用,从而起到保护肾功能的作用。 Objective: To analyse the tonifying kidney and promoting blood circulation prescription's inhibition of hyperirritable oxidation of the diabetic rats kidney and pancreas tissue. Methods: The SD rats are divided into 4 groups: natural comparison group( A group) , small dose group (B group), great dose group (C group ), DM group (D group). B, C,D groups arc formed DM model and administered the tonifying kidney and promoting blood circulation prescription( B group: 18g/kg; C group: 26g/kg). After 6 weeks, executing the rats by breaking their necks and getting their left kidneys and pancreata immediately to be formed into 10% thick fluid with physiological saline. Mensurating eoncentration of MDA,GSH - Px and SOD in kidney and pancreas tissue. Results The diabetic rat's concentration of MDA increases evidently and their concentration of GSH - Px, SOD decrcases evidently. The tonifying kidney and promoting blood circulation prescription can increase concentration of GSH - Px and SOD in kidney tissue and GSH - Px in pancreas tissue. Conclusion : The prescription has antioxidation to protect the kidney function through increasing concentration of GSH - Px and SOD in kidney tissue and GSH -Px in oancrcas tissue.
