烟草双特异性激酶基因(NtDSK2)的克隆与表达分析

摘要 利用对表达序列标签(expressed sequence tag,EST)数据库进行比对和搜索分析,并通过RT-PCR的方法从烟草中克隆到一个包含1218bp开放阅读框的新基因NtDSK2。NtDSK2编码了具有典型的双特异性激酶特征的蛋白。此蛋白与花生的STY激酶属于双特异性激酶的同一个亚家族。基于EST的数字化组织表达特征分析推测NtDSK2在烟草组织中广泛表达。运用半定量RT-PCR方法检测了烟草受烟草花叶病毒(tobacco mosaic virus,TMV)侵染后的表达特征。结果表明NtDSK2的表达量在TMV处理后随时间稳步上升,并在48h达到最大。推测NtDSK2是参与烟草抗胁迫调控的重要激酶基因。 A new gene named NtDSK2 was cloned in the research. NtDSK2 encoded a putative protein with 404 aa residues, which were characterized with STY domain. Expression characteristics were observed by semi-qualitative PCR after TMV infections. Results showed that expressions of NtDSK2 accumulated steadily and reached to its peak at 48h, after infection, which suggested that NtDSK2 may be an important kinase in the process of resistance to TMV.
