摘要
目的:用全脊髓匀浆(粗制髓鞘碱性蛋白)建立豚鼠实验性自身免疫性脑脊髓炎(EAE)模型。方法:Wistar大鼠全脊髓匀浆+完全福氏完全佐剂(CFA)免疫豚鼠,观察豚鼠的临床症状和中枢神经系统(CNS)的病理改变。结果:实验豚鼠均表现典型的EAE症状;光镜观察见CNS血管周围炎性细胞浸润和白质脱髓鞘改变。结论:全脊髓匀浆加完全福氏完全佐剂成功的诱导了豚鼠的EAE模型,具有模型稳定、发病率高、动物廉价易得的优点,是研究多发性硬化理想的动物模型。
Objective:To establish the experimental autoimmune encephalomyelitis (EAE) model on Guinea Pig with pan-spinal cord homogenate (coarse myelin basic protein,cMBP).Methods:The Guinea Pig was immunized with Wistar rat pan-spinal cord homogenate +Freund's complete adjuvant(CFA).The clinical symptoms and histopathologic changes of central nervous system were observed.Results:The Guinea Pigs in experimental group all demonstrated the typical symptoms of EAE with an incidence of 100%. The inflammatory cuff around small blood vessels, perivascular inflammatory cell infiltration and demyelination in white matter were observed through optical microscope.Conclusion:The EAE model of Guinea Pig was induced successfully ,the model was stable and with high incidence; besides the Guinea Pig was cheap and easy to get.Thus this model might be an ideal animal model to study multiple sclerosis.
