摘要 多药耐药(multidrug resistence,MDR)是肿瘤化疗失败的主要原因,多年来学者一直致力于多药耐药机制的研究。神经酰胺作为细胞凋亡的第二信使,参与多种细胞凋亡,它与凋亡相关因子bcl-2、NF-κB的关系一直是研究的重点。葡萄糖神经酰胺合成酶(glucosylceramide synthase,GCS)可催化神经酰胺糖基化使其转变为葡萄糖神经酰胺,许多研究证明GCS与MDR表型有关。 Multidrug resistance is considered to be the main obstacle to the effective chemotherapeutics, which has been investigated for years in the medical field. Ceramide functions as a second messenger to several signaling pathways, and the relationship between it and bcl-2 or NF-r,B has always been emphasized. Glucosylceramide synthase(GCS)catalyzes the glycosylation and inactivation of ceramide to glucosylceramide, which has been proved to be responsible for MDR phenotype.
