摘要
目的探讨干扰素γ对Ox-LDL刺激的巨噬细胞清道夫受体CD36表达的影响。方法收集小鼠腹腔巨噬细胞,培养后将贴壁细胞分为5组:1、正常对照组,用生理盐水刺激培养的巨噬细胞;2、Ox-LDL处理组,Ox-LDL 50mg/L刺激培养的巨噬细胞;3、4、5组为不同剂量的干扰素γ处理组,分别用不同剂量的重组INFγ(终末浓度分别为50、100、200ng/mL)刺激培养的巨噬细胞,然后加入Ox-LDL 50mg/L。24h后收集培养的细胞,用流式细胞仪测定巨噬细胞清道夫受体CD36的表达。结果Ox-LDL处理组与对照组比较,巨噬细胞清道夫受体CD36表达水平显著增高(P<0.01),干扰素γ各剂量组与Ox-LDL处理组比较,CD36表达显著减少(P<0.05)。结论Ox-LDL能明显刺激巨噬细胞清道夫受体CD36表达,干扰素γ抑制Ox-LDL引起的巨噬细胞清道夫受体CD36表达。
Objective To explore the effect of INF γ on the expression ofmacrophage CD36.Methods Mouse peritoneal macrophages were collected.After culturing,macrophages were stimulated by Ox-LDL and different doses of recombinate INF γ.Adherent cells were divided into 5 groups, l control group:macrophages were stimulated by normal saline.20x-LDL group:macrophages were stimulated by Ox-LDL(50mg/L).3 Recombinate INF γ groups:macrophages were stimulated by Ox-LDL 50 mg/L and recombinate INF γ( 50,100,200ng/mL).24 hours later, the expression of CD36 in macrophages by flow cytometry were detected.Results When macrophages were stimulated by Ox-LDL, CD36 expression increased significantly(P〈0.01).INF γ antagonized the expression of CD36 induced by Ox-LDL (INF γ groups vs Ox-LDL groups, P〈0.05).Conclusion Ox-LDL raised the expression of macrophages CD36. INF γ decreased the expression of CD36 induced by Ox-LDL.
