摘要
目的探讨中国汉族人群阿司匹林抵抗的发生与血小板膜糖蛋白(glycoprotein,GP)Ⅰa 807C/T多态性之间的关系。方法筛选200例动脉粥样硬化高危患者服用阿司匹林(100mg/d)至少7d以上,用二磷酸腺苷(ADP)和花生四烯酸(AA)作诱导剂测定血小板聚集功能,分为阿司匹林抵抗(AR)组、阿司匹林半抵抗(ASR)组,阿司匹林敏感(AS)组。并应用聚合酶链反应-序列特异性引物(polymerase chain reaction-sequence specific primers,PCR-SSP)方法检测各组GPⅠa 807C/T多态性。结果AR组,ASR组的T等位基因频率均显著高于AS组(P<0.005);AR组,ASR组的(TC+TT)基因型的频率也均高于AS组(P<0.05)。Logistic回归分析显示血小板膜GPⅠa T807等位基因与阿司匹林抵抗的发生显著相关(OR=3.76,95%CI:2.87~9.58)。结论血小板膜GPⅠa T807等位基因与阿司匹林抵抗的发生相关联,可能为阿司匹林抵抗发生的一种遗传易感性标志。
Objective To explore the correlation between the C807T dimorphism of platelet membrane GP I a gene and aspirin resistance.Methods A total of 200 patients with high risk of artherosclerosis took aspirin(100mg/ d) for more than 7 days,and we detected platelet aggregation function using adenosine diphosphate (ADP) and arachidonic acid(AA).The 200 patients were divided into three groups:aspirin resistant (AR) group,aspirin semiresponder(ASR)group and aspirin-sensitive(AS) group. Platelet GP I a gene 807CT polymorphisms were checked by polymerase chain reaction-sequence specific primers. Results T alleles in AR group and ASR group were higher than AS group (P〈0.005).The prevalence of genotypes(TT+TC) in AR and ASR group were significantly higher than that in AS group(P〈0.05).Platelet GP I T allele was significantly associated with aspirin resistance by multiple logistic regression(OR=3.76,95 % CI: 2.87-9.58). Conclusion The above data suggest that inherited platelet GP I a variations might have an important impact on aspirin resistance. The presence of GP I a T allele may be a marker of genetic susceptibility to aspirin resistance.
