摘要
目的观察补体C3水解片段C3f和去精氨酸C3f(C3f-des-arginine,DRC3f)对皮肤成纤维细胞合成与分泌转化生长因子-β1(transforming growth factor-β1,TGF-β1)的影响。方法采用人工合成的C3f(17肽)和DRC3f(16肽)刺激培养的正常成人皮肤成纤维细胞(NHDF-Ad),通过半定量逆转录多聚酶链式反应方法(RT-PCR)和酶联免疫法(ELISA)从mRNA和蛋白水平检测TGF-β1生成量。结果培养的正常成纤维细胞生成极微量TGF-β1(8 pg/ml)。C3f和DRC3f能够提高皮肤成纤维细胞TGF-β1生成量(P<0.05),且呈剂量依赖性。结论补体C3水解片段C3f和DRC3f提高皮肤成纤维细胞TGF-β1生成量的作用,可能对皮肤胶原纤维过度沉积起着促进作用。
Objective The increase of Complement C3f -dse -arginine (DRC3f) was found in systemic sclerosis (SSc) in our study recently. This report is aimed to investigate the influence of synthesized C3f and DRC3f on production of TGF - β1 by normal human adult dermal fibroblasts (NHDF- Ad). Methods The cultured NHDF- Ad was stimulated with synthesized C3f and DRC3f,the TGF -61 mRNA in the cells was detected by semi- quantitative reverse -transcriptional PCR ( RT - PCR), while the TGF - β1 molecules in the supernatant of the cells were measured by enzyme linked immunosorbent assay (ELISA). Results The cultured NHDF - Ad only produced trace of TGF -β1 (7 ng/ml). After stimulation by C3f or DRC3f, the expression of TGF - β1 at both mRNA and protein levels in NHDF - Ad was increased significantly in dose - dependent manner. Conclusions These results suggest that the up - regulation of TGF - βin NHDF-Ad by C3f and DRC3f may participate in the type Ⅰ collagen over- expression and deposit in skin of patients with SSc.
