针刺联合紫杉醇对小鼠Lewis肺癌细胞凋亡相关因子p53、bcl-2蛋白表达的影响

Effect of Acupuncture Combined with TAX on the Expression of Apoptosis Related Factor p53 and bcl-2 of Mice Lewis Lung Cancer Cells

目的本实验采用针刺联合抗癌新药紫杉醇对Lewis肺癌小鼠进行治疗,探讨针药结合对肿瘤的治疗作用及机制。方法Lewis肺癌移植瘤小鼠48只,体重18～22g,随机分为4组,即模型组、针刺组、药物组、针药结合组,每组12只。观察小鼠的生存状态、肿瘤体积的动态变化,应用western Blotting法观察肿瘤组织中凋亡相关因子p53、Bc1-2蛋白表达情况.结果针刺、紫杉醇、针剌联合紫杉醇均有抑瘤作用,但针药结合抑瘤作用最强,且能改善实验动物的生存状态.各组中均有p53、bc1-2蛋白表达,模型组表达最高;与模型组比较,针刺组、药物组、针药结合组表达明显低于模型组,有显著差异;针药结合组与针刺组和药物组比较,有显著差异.结论针剌加紫杉醇具有联合抑制野生型p53基因突变,降低bc1-2蛋白表达的作用,两者具有协同效应。

Objective We adopt acupuncture associating with TAX, to investigate the anti-cancer effect and mechanism. Metheds 48 mice with inoculated Lewis Lung Carcinoma were divided randomly into 4 groups： model group, acupuncture group, medicine group, combination of acupuncture and medicine group. Growth state of the mice and change curve of tumour. Protein expression of apoptosis related factor p53 and Bcl-2 in turnout were observed by Western Blotting method. Results Acupuncture, TAX and the combination of acupuncture and TAX separately had anti-tumor effect, with the combination was the best arid improve growth state of the mice. p53 and bcl-2 can be detected in four groups. Compared with model group, the expression of other groups was lower. Compared with acupuncture group and medicine group, the exprission of combination group of acupuncture and TAX was lower. Conclusions Combination of acupuncture and TAX can prohibit p53 Mutant and decrease Bcl-2, which have co-operating effect.
