细菌碱性蛋白酶促大豆分离蛋白聚集

摘要 研究了大豆分离蛋白(SPI)的细菌碱性蛋白酶水解产物的聚集行为。采用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)和高效分子排阻色谱分析了细菌碱性蛋白酶对SPI的降解模式,结果显示,降解产物大部分为分子质量小于6.5kDa的小分子量组分,反应过程有不明聚集物生成。浊度分析表明,此聚集物可能由小分子的降解产物组成。在较高的蛋白质浓度下,蛋白酶甚至可以催化SPI形成弹性凝胶。 The objective of this study was to investigate the aggregation of soy protein isolates (SPI) induced by Protex. SDS-PAGE and SE-HPLC analyses showed that the SPI proteins were digested to small fragments with molecular weights less than 60.5kDa, and unknown aggregates were generated in the solution. The aggregates were formed by the major fragments according to the turbidity analysis. The Protex was able to induce gelation of SPI at high concentration.
