摘要
对建立高效毛细管电泳方法检测蛋白质进行研究,探讨了在碱性缓冲溶液中添加SiO2纳米材料对人血清白蛋白(HSA)检测的影响.实验发现:采用未处理过的75μm(内径)×60 cm(50 cm有效长度)毛细管柱、15 kV分离电压、25 mmol.L-1硼酸硼砂组成的缓冲溶液直接检测HSA,由于HSA在毛细管内表面产生强烈吸附而无法检测到相应的电泳峰;上述缓冲溶液中加入一定量的纳米材料后,在电泳图中8 min左右出现尖锐的HSA峰,该峰峰形良好,完全可以达到检测与定量的要求.该方法对HSA的检测限为0.50 mg.L-1,HSA峰迁移时间的相对标准偏差为2.62%.实验结果表明SiO2纳米材料是检测蛋白质的有效添加剂.
The aim of this work is to detect Human Serum Albumin by capillary electrophoresis (CE) with SiO2 nanoparticles (SNPs) as additives in basic buffer. The CE experiments are carried out in 75 μm× 60 cm (the effective length is 50 era) bare silica capillary filled with 25 mmol·L^-1 borate buffer cross which is applied a voltage of 15 kV. It is found that in the absence of nanoparticles, the proteins could not be detected due to their strong adsorption onto the silica wall. However, sharp peaks are observed at around 8 min in the presence of silica nanoparticles. The peak can be quantified with 0. 5 mg·L^-1 in detection limit and 2.62% in relative standard deviation of migration time.
