尿毒症脑病大鼠脑组织谷氨酸和γ-氨基丁酸浓度变化

Concentration variations of glutamate and γ-aminobutyric acid in brain tissues of uremia encephalopathy rat

目的研究尿毒症脑病大鼠大脑组织中谷氨酸(Glu)和γ-氨基丁酸(GABA)浓度变化与尿毒症脑病发病的相关性。方法Wister大鼠36只,随机分为两组:正常对照组(12只)、模型组(24只)。模型组大鼠给予顺铂诱导急性肾功能衰竭,再将模型组大鼠分为尿毒症组和尿毒症脑病组。高效液相色谱法分别检测上述三组大鼠大脑组织内Glu和GABA的含量。结果尿毒症组和尿毒症脑病组大鼠大脑组织中Glu和GABA浓度发生变化,尿毒症组大鼠大脑组织中Glu和GABA浓度与正常组相比,差别没有统计学意义(P>0.05)。尿毒症脑病大鼠大脑组织中Glu和GABA浓度与正常组和尿毒症组相比,差别均有统计学意义(P<0.01)。结论急性肾功能衰竭并发脑病时,脑组织中Glu和GABA浓度发生明显变化,因此Glu和GABA可能参与了尿毒症脑病的发生。

Objective To detect concentration variations of Glutamate and γ-aminobutyric acid in brain tissues of acute renal failure rats with uremia encephalopathy and understand the relationship with uremia encephalopathy, Methods Thirty-six male healthy Wister rats were divided into 2 groups by random.. 12 in normal control group, 24 in model group. In the model group, Cisplatin was administered to induce acute renal failure. The model group was subdivided into uremia group and uremia encephalopathy group. High performance liquid chromatography （HPLC） was used to detect Glutamate γ-aminobutyric acid contents in rat brain tissues of the 3 groups. Results Concentrations of glutamate and γ-aminobutyric acid in rat brain tissues of uremia group and uremia encephalopathy group changed. Compared with normal control group, the concentration of Glutamate γ-aminobutyric acid in uremia group showed no significant difference （P〉0. 05）. Compared with normal control group and uremia group, uremia encephalopathy group showed significant difference （P 〈 0. 01）. Conclusion The contents of Glutamate and γ-aminobutyric acid were significantly changed when acute renal failure with uremia encephalopathy occurs. So Glutamate γ and aminobutyric acid may play a great role in the mechanism of uremia encephalopathy.
