赤芍和茜草水提取物对系统性硬皮病成纤维细胞增殖及胶原合成影响的研究 被引量:6

摘要 目的:研究赤芍和茜草水提取物对系统性硬皮病成纤维细胞增殖和I、Ⅲ型前胶原mRNA合成的影响。方法:MTT法测定对成纤维细胞增殖的影响,RT-PCR测定对成纤维细胞Ⅰ、Ⅲ型胶原mRNA合成的影响。结果:赤芍和茜草水提取物对成纤维细胞的增殖和Ⅰ、Ⅲ型前胶原mRNA表达都具有一定的抑制作用。赤芍水提取物可以使Ⅰ型前胶原表达减少29%,Ⅲ型减少33%;茜草水提取物使Ⅰ型减少27%,Ⅲ型减少23%。结论:赤芍和茜草水提取物能够抑制系统性硬皮病成纤维细胞增殖,不同程度的抑制Ⅰ、Ⅲ型胶原mRNA的表达,具有一定的抗纤维化作用。 Objectives: To study effects of the aqueous extracts of Red Peony Root (RPR) and India Madder Root(IMR) on proliferation and procollagen mRNA synthesis of skin fibroblasts from systemic sclerosis patients. Methods. MTT test was used to study cell proliferation and procollagen mRNA expression was studied by RT - PCR. Results. Both the aqueous extracts of RPR and IMR suppressed cell proliferation and inhibited the expression of type Ⅰ and Ⅲ procollagen mRNA of fibroblasts from systemic sclerosis patients. The aqueous extracts of RPR reduced the expresstion of procollagen Ⅰ mRNA by 29 %, procollagen Ⅲ mRNA by 33 %. The aqueous extracts of IMR decreased procollagen Ⅰ mRNA by 27 %, procollagen Ⅲ mRNA by 23 %. Conclusions: Both the aqueous extracts of RPR and IMR suppressed the fibroblasts proliferation and expression of procollagen mRNA. This may be the molecular pharmacodynamics of them on treatment of SSc.
