云南布朗族群体15个STR基因座遗传多态性研究 被引量:6

摘要 目的采用PCR-STR及基因分型技术,探讨云南地区布朗族群体中D3S1358、TH01等15个常染色体基因座的遗传多态性,建立布朗族群体的遗传学基础数据,为法医学应用提供依据.方法采集97名云南地区布朗族无关个体的静脉血,经典有机溶剂法提取DNA,应用PCR技术,复合扩增D3S1358、TH01等15个常染色体基因座的短串连重复序列,产物用遗传分析仪电泳荧光检测,调查云南地区布朗族15个STR基因座的等位基因频率,进行遗传多态性分析.结果15个STR基因座在云南布朗族群体中具有遗传多态性,基因型分布符合Hardy-Weinberg平衡定律(P>0.05).其累积匹配概率(Pm)为7.25×10-16,累积非父排除率(CPE)为99.9999966%,累积个人识别能力(CDP)为99.9999999999999%.结论云南布朗族群体15个常染色体STR基因座有较高的非父排除率和个体识别能力,可为法医学亲子鉴定和个体识别及遗传学研究提供依据. Objective To explore the short tandem repeat (STR) loci polymorphism in Yunnan Bulang nationality to provide basic data for population study, forensic cases and parentage testing. Methods The genetic distributions of 15 STR loci were investigated with the use of co - amplification, genescan and genotype in blood samples obtained from 97 unrelated Bulang nationality. Results 118 STR alleles and 229 genotype were obtained in 15 loci, with the allele frequencies ranging from 0. 005 2 -0. 556 7, heterozygosity (H) 0. 556 7 - 0. 876 3; discrimination power ( DP ) 0. 745 9 - 0.9565, cumulative discrimination power ( CDP ) 0. 999 999 999 999 999% , probability of exclusion ( PE ) 0. 354 7 - 0. 841 7, cumulative probability of exclusion (CPE) 0. 999 999 966% ; polymorphism information content ( PIC ) 0. 502 8 - 0. 852 5. Conclusions The 15 STR loci used in this study is highly polymorphic in Bulang population and can be applied to population study and forensic practice.
