摘要
目的:探讨自然临产及缩宫素诱导临产(OT引产)子宫体部平滑肌基因表达谱的差异。方法:应用8064条人类全长基因的cDNA表达谱芯片,分别筛选自然临产与OT引产活跃期比未临产子宫体部平滑肌组织的差异基因表达谱,并用RT-PCR验证HSPA1B基因的表达。结果:(1)分娩活跃期子宫体部与未临产相比,显著性差异表达基因,自然临产时有469条,OT引产时有539条。二者差异基因表达谱中相同表达趋势的基因为338,占总差异基因数的54%,但多数基因在OT引产时上调的比值高于自然临产;(2)HSPA1B基因在未临产组、自然临产组及OT引产组子宫体部的相对表达量分别为0.40±0.39、0.80±0.75和1.11±0.78。自然临产组及OT引产组均明显高于未临产组(P<0.05),与芯片实验结果一致。结论:自然临产与OT引产子宫体部平滑肌组织差异基因表达谱具有相似性,但OT引产时多数基因上调的比值高于自然临产,对共同差异表达基因的深入研究,可能为揭示分娩动因、监测产程和设计相关药物提供新的思路。
Objective:To investigate the differentially expressed genes in corpus myometrium from spontaneous labor and oxytocin induced-labor( OT labor) at active stage. Methods: eDNA microarray, with 8 064 unique probes, was used to comprehensively scan the differentially expressed genes from corpus myometrium in spontaneous or OT labor verse those not in labor(no-labor) respectively. Expression of heat shock 70kD protein 1B ( HSPA1B ) was confirmed using RT-PCR. Results: ( 1 )469 and 539 genes were found to be differentially expressed in corpus myometrium from spontaneous or OT labor verse no-labor,respectively ,338 differentially expressed genes (54% in the genes) were consistent. The up-regulated ratio of genes at OT labor was higher than at spontaneous labor; (2)The relative expression of HSPA1B in no-labor, spontaneous labor and OT labor were 0.40 4- 0.39,0.80 4- 0.75 and 1.11 4- 0.78, respectively. There was remarkable difference in HSPA1B between spontaneous or OT labor and no-labor( P 〈0.05). The results of eDNA microarray and RT-PCR technology were strongly confirmed. Conclusion:Similar gene profiling between spontaneous and OT labor is observed, but differentially expressed ratio of most genes in OT labor was higher than that in spontaneous labor. Our study may help for disclosing onset of labor, monitoring delivery and designing relative drugs.
