摘要
目的观察丹红注射液对动脉粥样硬化(AS)家兔模型脂代谢及血管内皮功能的影响,探讨丹红注射液抗AS作用及可能机制。方法36只新西兰雄性白兔随机均分为正常对照组、高胆固醇组(HC组)、高胆固醇加氟伐他汀组(FC组)和高胆固醇加丹红注射液组(DC组);组织形态学分析AS斑块/内膜面积比值及斑块最厚处内膜厚度/中膜厚度比值;酶法检测血脂;酶法测定血清一氧化氮(NO)和血浆内皮素(ET)水平。结果DC组与HC组比较,血清TC、LDL-C明显降低(P<0.05);血管AS病变显著减轻(P<0.05),血清NO水平显著增高(P<0.05);血浆ET水平显著降低(P<0.05)。结论丹红注射液对实验性AS具有抑制作用,其作用机制可能为①降低血清TC和LDL-C水平,延缓AS斑块的形成;②调节血管内皮细胞生成和释放NO、ET,保护血管内皮细胞功能,进而产生抗AS作用。
Objective:To observe the effects of Danhong injection on lipid metabolism and vascular endothelial function in rabbit model with experimental atherosclerosis and explore its possible mechanism. Method: Thirty-six New Zealand male rabbits were randomly divided into normal, control, group (n= 9), high-cholesterol, group (HC group, n= 9), high-cholesterol plus fluvastatin (FC group, n=9) and high-cholesterol plus Danhong injection group(DC group, n= 9) and then were fed for twelve weeks. The plaque area of aorta and the neointima thickness were performed with histomorphometry analysis. Serum lipids, serum NO, plasma ET were detected with standard enzymatic assays. Result:There was significantly difference in the serum lipids TC and LDL-C between the HC group and DC group(P〈0. 05), the neointima thickness and plaque area of aorta and the level of serum NO in the DC group were significantly decreased compared with the HC group(P〈0.05), but the level of plasma ET increased. Conclusion: The study suggests that Danhong injection could decrease atherosclerosis in atherosclerotic rabbit model by decreasing the serum lipids TC and LDL-C levels and releasing the NO and ET.
