摘要 目的研究人外周血干细胞(PBSC)向血管内皮细胞分化。方法以血管内皮细胞生长因子(VEGF)和碱性成纤维细胞生长因子(bFGF)对经重组人粒细胞集落刺激因子刺激后采集的PBSC进行诱导培养,观察细胞形态的变化,运用流式细胞术检测诱导后的细胞表达CD31和vWF情况,并用透射电镜观察细胞的超微结构。结果经VEGF和bF-GF诱导后的细胞形态上表现内皮细胞特征,经流式细胞仪测定表达血管内皮细胞特有表面标志CD31和vWF,透射电镜细胞胞浆内可见特征性Weible-Palade小体。结论外周血干细胞在VEGF和bFGF诱导下,可分化为内皮细胞。 Objective To study differentiation of human peripheral blood stem cells (PBSC) into blood vessel endothelial cells. Methods The PBSCs were collected after stimulation of granulocyte-colony stimulating factor(G-CSF) and then cultured and induced by vascular endothelial growth factor(VEGF) and basic fibroblast growth factor( bFGF) in vitro. The morphological changes of the cells were observed. The cells were enumerated by flow cytometry (FCM) with CD31 monoclonal antibody and vWF monoclonal antibody, and their ultrastructural characteristics were observed by electronic microscopy. Results After induced by VEGF and bFGF in vitro, PBSCs became endothelial cell like cells and CD31 and vWF were expressed. The Weible-Palade bodies were demonstrated in the cells under the observation of electronic microscopy. Conclusion Peripheral blood stem cells can differentiate into endothelial cells by the induction of VEGF and bFGF.
