摘要 目的探讨As2O3对NB4细胞PTTG和c-myc基因表达的影响。方法用RT-PCR的方法检测As2O3对NB4细胞PTTG和c-myc的表达的影响。结果0.5、1.0、2.0μmo1/L的As2O3作用NB4细胞24、48h后PTTG基因的表达强度逐渐减弱;与未加As2O3的NB4细胞对照组比较P<0.05。c-myc基因的表达强度亦逐渐减弱,与未加As2O3干预的NB4对照组比较P<0.05。结论PTTG和c-myc在NB4细胞中也有高表达,提示PTTG和c-myc的高表达在急性早幼粒细胞白血病(APL)发病过程中可能起着重要作用,As2O3能有效地下调NB4细胞PTTG和c-myc的表达。 Objective To investigate the effects of arsenious acid( As203 ) on the expression of PTTG and c-myc in NB4 cell line. Methods The expression of PTTG and c-myc gene mRNA was determined by reverse transcriptasepolymerase chain reaction(RT-PCR). Results After 0. 5,1.0,2.01μmol/L As203 treatment for 24,48 h, the expression of PTTG and c-myc of NB4 cell decreased, P 〈 0. 05 vs control. The decrease in the expression of PTTG and c-myc in NB4 ceil line was negatively correlated with the dose and duration of action of As2O3. Conclusion In vitro,PTrG and c-myc are expressed high in NB4 cell. Overexpression of PTTG and c-myc may be related to the progression of APL. As2O3 can significantly decrease the expression of PTTG and c-myc of NB4 cells.
