积雪草离体培养和快速繁殖方法探讨 被引量:4

摘要 【目的】探讨采用植物组织培养技术进行积雪草快速繁殖的方法,为积雪草的人工栽培及优良品种选育奠定基础。【方法】以积雪草的带节茎为材料,研究离体培养与植株再生的过程。【结果】Murashge and Tucker(MT)基本培养基加N-苯基-N-′1,-2,-3-噻二唑-5-脲(TDZ)0.5 mg/L,有利于丛生芽的产生,不定芽诱导率为87.6%;根的诱导以MT+吲哚乙酸(IAA)0.2 mg/L培养基较好,生根率为100%,且植株生长旺盛。试管苗移栽,成活率可达90%左右。【结论】在MT培养基中加入TDZ可增加积雪草带节茎组织培养的成芽率,移入添加0.2 mg/L IAA的MT培养基中培养可增加其生根率,该方法为积雪草工厂化离体繁殖奠定了基础。 [Objective] To find out technological evidence for the rapid propagation of Centella asiatica (L.) Urb.. [Methods] The nodular stem segment of Centella asiatica (L.) Urb. was taken as the explant to induce fascicular bud. [ Results] MT culture medium with 0.5 mg/L thidiazuron (TDZ) added was effective to induce the bud formation, and the shooting rate was 87.6%. As for the induction of rooting, MT culture medium with 0.2 mg/L indoleacetic acid (LAA) added was optimal, the rooting rate being 100% and the plant growing well. The survival rate of the transplanted test-tube plantlet arrived 90%. [ Conclusion] MT culture medium with TDZ added can increase the shooting rate of nodular stem segment of Centella asiatica ( L. ) Urb., and MT culture medium with 0.2 mg/L IAA can increase the rooting rate of the bud, indicating that this method can serve as the preliminary in-vitro propagation system for Centella asiatica (L.) Urb..
