大鼠脑组织腺苷含量的HPLC分析

摘要 用高效液相色谱(HPLC)配紫外检测器,分离测定大鼠脑组织中腺苷含量。经实验确定以甲醇(10%~62%)、磷酸缓冲液(90%~38%)为流动相梯度洗脱,样品液中腺苷含量在2mg/L~10mg/L范围内线性关系良好。加入标准样品的平均回收率为(966±20)%。对6mg/L样品10次重复测定结果的标准偏差为014,变异系数为31%。对大鼠脑组织样品测定结果发现:与3~6月龄成龄鼠比较,18~20月龄老龄大鼠大脑皮层、海马、纹状体中腺苷含量分别增加了667%、372%和273%,不同脑区腺苷含量也有明显差异。结果提示不同脑区腺苷含量的随龄增加可作为研究衰老的一项指标; Using HPLC combine with a UV detector to separate and identify Adenosine content in rat brain,it was found that Methanol (10%~62%) and Phosphate buffer (90%~62%) as a motive phase for gradient elution,the content of Adenosine in the sample at 2mg/L~10mg/L gave a good linear relationship.The everage recovery rate of standard Adenosine added to the sample was (96 6±2 0)%.The sample of 6mg/L was identified.Through 10 times identification,its standard deviation ( SD ) was 0.14.The coefficient of variability was 3 1%.The experiment of comparison between 3~6 months and 18~20 months rats showed that the Adenosine contents in brain cortex,hippocampus and striatum raised 66 7%,37 2% and 27 3% respectively.Adenosine content of different regions were obviously different.The results showed that the increasing Adenosine content of different brain region according to ages could be as indicator for investigating ageing.And the inhibition of the release of acetylcholine (Ach) by ageing accumulation of Adenosine in contral nervous system could be one of the important reason of memory loss of senile functional problems.
