摘要 目的:检测肾癌原发灶和转移灶中KISS-1基因的表达并在体外实验中验证其缺失对细胞侵袭能力的影响。方法:选择30例肾癌患者,应用荧光定量PCR检测肾癌原发灶和转移灶中KISS-1基因mRNA的表达,应用RNA干扰技术获得KISS-1基因表达缺失的肾癌细胞株,检测干扰前后细胞的侵袭能力变化。结果:转移灶中的KISS-1mRNA表达较原发灶中明显下调,KISS-1表达与肿瘤远处转移呈显著相关。干涉前后都有细胞穿透滤膜,干涉后细胞数明显高于干涉前,细胞数分别为31±6和78±8,两组有显著差异(P<0.05)。KISS-1基因表达缺失的肾癌细胞侵袭能力明显增强。结论:KISS-1基因在肾癌中具有转移抑制作用,可能在肾癌转移治疗中发挥作用。 Objective: To detemtine the expression of KISS- 1 gene, one of metastasis suppressor genes which play a critical role in tumor invasion and metastasis,in primary and metastatic renal cell carcinoma,and to study the effect of KISS-1 gene depletion on the invasion of tumor cell in vitro. Methods:The expression of KISS-1 mRNA in 30 patients with primary and metastatic renal cell carcinoma was detected by fluorescence quantitative polymerase chain reaction. The renal cell carcinoma cell line with KISS-1 gene depletion was prepared by RNA interference,and the changes in the invasion ability were measured. Results:The expression of KISS-1 mRNA was much lower in the metastatic tumor than in the primary tumor and was significantly correlated with tumor metastasis. The cells penetrated the filter membrane both before and after RNA interference,the cell number after RNA interference was significantly higher than that before RNA interference (78+8 vs. 31±6,P〈 0.05). Conclusion:KISS-1 gene could inhibit the invasion ablility of renal cell carcinoma and is expected to be useful in antimetastatic therapy of renal cell carcinoma.
