摘要
目的:探讨鼠双微体基因(MDM2)表达与不同人类乳头状瘤病毒(HPV)感染状态的外阴鳞癌发生、发展的关系。方法:采用RT-PCR和Western blotting检测42例外阴鳞癌组织、10例正常外阴组织中MDM2基因mRNA及蛋白表达,PCR检测其HPV-DNA,分析HPV阳性组和阴性组外阴鳞癌组织中MDM2基因mRNA及蛋白表达与其临床病理特征的关系。结果:MDM2基因mRNA在外阴鳞癌组织中相对含量及阳性表达率(0.54±0.35,38.1%)明显高于正常外阴组织(0.21±0.04,0;P<0.05);低分化癌(1.01±0.45,72.7%)与中、高分化癌(0.41±0.18,27.8%;0.32±0.03,23.1%)分别比较,差异有显著性(P<0.05)。MDM2蛋白在外阴鳞癌组织中相对含量及阳性表达率(1.11±0.16,33.3%)明显高于正常外阴组织(0.87±0.02,0;P<0.05);低分化癌(1.27±0.14,63.6%)与中、高分化癌(1.09±0.17,22.2%;1.04±0.12,23.1%)分别比较,差异有显著性(P<0.05)。MDM2基因mRNA及蛋白表达在HPV阳性组与阴性组组间比较,差异无显著性。结论:在外阴鳞癌的形成和发展中MDM2基因与HPV可能作为两个独立的因素起作用;MDM2可能参与了外阴鳞癌的发生发展过程,并可能成为与外阴鳞癌发生发展相关的癌基因。
Objective:To investigate the relationship between MDM2 expression and human papillomavirus (HPV) status in vulvar squa- mous cell carcinomas (VSCC). Methods:The expressions of MDM2 mRNA,MDM2 protein,and HPV DNA in 42 cases of VSCC and 10 normal vulvar tissues were determined by reverse transcription polymerase chain reaction (RT-PCR), Western blotting,and PCR,respectively. Results:The relative content and positive expression rate of MDM2 mRNA in VSCC were significantly higher than those in normal vulvar tissues (0.54±0.35 vs. 0.21±0.04,38.1% vs. 0,P 〈 0.05 ). The relative content and positive expression rate of MDM2 mRNA were 1.01± 0.45 and 72.7%,0.41±0.18 and 27.8%,and 0.32±0.03 and 23.1% in poorly,moderately,and highly differentiated VSCC,respectively,and there were significant differences between poorly differentiated VSCC and moderately and highly differentiated VSCC (P 〈 0.05 ). The relative content and positive expression rate of MDM2 protein in VSCC were significantly higher than those in normal vulvar tissues (1.11±0.16 vs. 0.87±0.02,33.3% vs. 0,P 〈 0.05 ). The relative content and positive expression rate of MDM2 protein were 1.27±0.14 and 63.6%, 1.09±0.17 and 22.2%,and 1.04±0.12 and 23.1% in poorly,moderately,and highly differentiated VSCC,respectively,and there were significant differences between pcody differentiated VSCC and moderately and highly differentiated VSCC (P 〈 0.05). No significant difference was found in the expressions of MDM2 mRNA and protein between HPV-positive group and HPV-negative group. Conclusion:MDM2 and HPV may be 2 independent factors during the formation and development of VSCC. MDM2 is involved in the occurrence and development of VSCC.
