摘要
目的:探讨失血性休克对下丘脑室旁核(PVN)促肾上腺皮质激素释放激素(CRH)和血管加压素(AVP)基因转录的影响。方法:大鼠分别于失血后0、15、30、60、120min断头;RIA法测血中激素水平;应用[35S]标识的RNA探针进行原位杂交实验,分析失血后PVN内CRH和AVP基因转录的动力学变化。结果:血中促肾上腺皮质激素(ACTH)及AVP于失血后15min达高峰(P<0.01),失血后60min时基本恢复到基础水平;PVN内CRHh nRNA在失血前几乎无表达,于失血后15min表达量已显著升高(P<0.01),30min达峰值(P<0.01),120min内基本恢复,失血前CRHmRNA在PVN就有大量表达,于失血后15min及120min两次有意义升高(P<0.01);失血前AVPhnRNA在PVN的小细胞领域基本没有表达,于失血后15min开始有意义增加(P<0.01),30min达峰值(P<0.01)后一直持续,失血前AVPmRNA在该领域的表达量很少,失血后60min时其表达量有意义增加(P<0.01),并持续到失血后120min;失血前AVPhnRNA在PVN的大细胞领域就有大量表达,于失血后15min开始显著升高(P<0.01),并一直持续,失血前AVPmRNA在该领域的表达量很多,失血后其表达量没有明显变化。结论:在失血性休克时共存于小细胞领域的CRH和AVP的基因转录的动态变化不同,分别存在于大小细胞两领域的AVP的基因转录的动态变化不同。
Objective: To investigate the effect of hemorrhagic shock on corticotropin releasing hormone (CRH) and arginine vasopressin (AVP) gene transcription in paraventricular nucleus (PAN) of hypothalamus. Methods:Rat model of hemorrhagic shock was established by drawing blood from the right atrium,and the rats were decapitated 15,30,60,and 120 minutes after hemorrhagic shock. In situ hybridization was performed by using [35S]-labeled riboprobes. Results:The plasma levels of ACTH and AVP increased rapidly,reached the peak at 15 minutes,and then gradually returned to basehne. CRH and AVP heteronuclear RNA (hnRNA) significantly increased at 15minutes. AVP hnRNA further increased at 30 minutes and maintained relatively stable until 120 minutes. The expression of A VP hnRNA in the magnocellular division increased at 15minutes and maintained high, but in the same division it was not different from that at baseline at any time. Conclusion:The dynamic changes of CRH and AVP gene transcription in the parvocellular division are different,and the dynamic changes of AVP gene transcription in the magnocellular division also differs from those in the parvocellular division.
