摘要 目的:建立人血浆中苯溴马隆浓度的HPLC测定方法。方法:色谱柱为Hypersil BDS C_(18)(250 mm×4.6 mm,5μm);流动相为甲醇.水.三乙胺(48:52:0.02),流速1ml·min^(-1);紫外检测波长357 nm;以香豆素为内标,采用乙腈直接沉淀蛋白后进样。结果:苯溴马隆的线性范围为0.01~0.5 mg·L^(-1)(r=0.999 2);提取回收率为103.5%,106.8%,方法回收率为109.5~113.2%,日内和日间RSD分别小于7.0%和9.5%结论:本法操作简便、灵敏、准确,可用于临床治疗度高。 Objective: To develop a method for the determination of Benzbromarone concentration in human plasma by HPLC. Method: The Hypersil BDS C,s column(250 mm× 4.6 mm, 5μm)was used. The mobile phase was methanol-water- triethylamine(48 :52: 0.02) and flow rate was 1 ml·min^-1. The detecting wavelength was at 357 nm. Cumarin was used as the internal standard. The sample was deposited by aeetonitrile and then injeetied. Result: The linear range of benzbromarone was 103.5% - 106.8%, The method recovery was 0.01 - 0.5 mg· L^-1 ( r = 0.999 9), the extration recovery was 109.5 - 113.2%, the with in day less than 7.0% and between day lessthan 9.5%. Conclusion: The method is simple, sensitive and accurate which is sumitable for clinic therapeutic drug monitoring.
