摘要
以辣根过氧化物酶(HRP)催化H2O2氧化邻苯二胺(OPD)为研究体系,利用紫外-可见分光光度法,以反应初始速率为测试参数,研究了缓冲溶液种类,酸度条件以及保存时间对HRP稳定性的影响,发现HRP在磷酸盐-磷酸二氢钾缓冲溶液中保存较稳定,最佳保存pH为8.0,0.1μg/mL HRP溶液保存7天活性未见明显变化,结果令人满意.
Effects of buffer solutions, acidic condition and conservation time on activity of horseradish peroxides (HRP) in Solutions were investigated by UV - visible spectroscopy. This study based on the reaction of hydrogen peroxide oxidized o - phenylenediamIne with HRP. The initial rate of the HRP - catalyzed reaction was selected as parameter for determination of the activity of HRP. The results showed that the horseradish peroxides had better stability within 7days in phosphate buffer solution and the optimal pH was 8.0. The result is satisfactory.
