西瓜和甜瓜杂种一代种子纯度的RAPD鉴定 被引量:18

摘要 选用100条随机引物(S71-S190)对西瓜、甜瓜杂种一代和其父、母本的种子进行随机扩增多态性DNA(RAPD)分析,探索西瓜和甜瓜杂交品种种子纯度鉴定的便捷途径.结果表明:在100条引物中筛选出4条有特征条带的引物,其中S92号引物能使甜籽一号、白兰蜜和黄河蜜6号与其母本有效区分开来;S162号、S181号引物能使甜籽一号与其母本区分开来;S175号引物能使白兰蜜与其母本区分开来;但未发现使T×14E与其母本区分开来的引物. The 100 random primers(S71 - S190)were tested in random amplified polymorphic DNA (RAPD) analysis for purity identification of F1 hybrid seeds and their parents in water melon and melon.The results showed that 4 primers with specific bands of RAPD profiles were selected from 100 random primers,among which S92 primer was available for differentiating seeds of F1 hybrid cultivars, such as ‘ Tianzi 1’ in water melon,‘ Bailanmi’ and ‘ Huanghemi 6’ in melon, from their female parents, S162 and S181 primers for the hybrid ‘ Bailanmi’ from its female parent,S175 primer for ‘ Huanghemi 6’ from its female parent. But no one primer was found out for distinguishing between the hybrid ‘T × 14E’ in water melon from its female parent. The result could be used as the basis of identification of hybrid cultivars in water melon and melon.
