丰花月季愈伤组织的诱导及细胞悬浮培养 被引量:15

摘要 分别以丰花月季叶片、叶柄和茎段为外植体诱导出愈伤组织,并建立细胞悬浮系。结果表明,丰花月季叶片和茎段是诱导愈伤组织和进行细胞悬浮培养的理想外植体;诱导松散型愈伤组织的最佳激素浓度配比是NAA 4 mg·L-1+2,4-D 1 mg.L-1+BA 1 mg.L-1;细胞悬浮培养细胞最佳起始密度为1.5×104~3.0×104个.mL-1时;继代培养4~5代时,可得到稳定的悬浮细胞系。 Cell suspension cultures were established from the callus tissues which were induced from leaf, leafstalk and stem segments of Rosa floribuncla. The result indicated that both leaf and stem explants were suitable for cell suspension. Friable callus was obtained when cultured on Murashige and Skoog (MS) medium supplemented with 4 mg·L^-1 NAA, 1 mg·L^-1 2, 4-dichlorophenoxyacetic acid (2,4-D), 1 mg·L^-1 6-benzyladenine (6-BA). The best origination density was 1.5×10^4 -3.0×10^4 number·mL^-1; after 4-5 generations of subculture, stable cell suspension was obtained.
