摘要
目的:观察外源性bFGF(basic fibroblast growth factor)对雏鸡形觉剥夺性近视眼(form deprivation myopia,FDM)眼球后壁组织血管活性肠肽(vasoactive intestinal peptide,VIP)表达的影响。方法:将出生2d龄的40只来亨雏鸡随机分为对照组和实验组,两组右眼为遮盖眼,左眼为自身对照眼。对照组遮盖眼注射1g/L牛血清白蛋白(bovine scrum albumin,BSA)溶液20μL,共10只鸡;实验组遮盖眼玻璃体腔注射bFGF1,2,4ng+1g/LBSA溶液20μL,每种剂量10只鸡。1wk后采用SP法免疫组织化学染色和逆转录聚合酶链反应检测眼球后壁组织VIP的表达。所有实验数据,对照组和实验组比较采用单因素方差分析,不同浓度实验组间采用SNK-q检验。结果:VIP的表达主要位于视网膜光感受器外节、内核层、内丛状层和脉络膜,在神经节细胞层和神经纤维层有较弱表达。所有组自身对照眼的VIP表达两两比较均无明显差异(P>0.05);实验组与对照组遮盖眼相比较差别有统计学意义,VIP表达在实验组明显上调(P<0.01),并且VIP的表达在实验组组间的差别有统计学意义,随药物剂量增加有逐渐上调的趋势(P<0.05)。结论:外源性bFGF可上调鸡FDM眼球后壁组织VIP的表达。
AIM: To investigate the exogenous basic fibroblast growth factor (bFGF) as a regulator on the expression of vasoactive intestinal peptide (VIP) on form deprivation myopia (FDM) of chick retinal-choroids-sclera.
METHODS: Forty two-day-old lehorn chickens were divided randomly into control group and experimental group. The right eyes of the chickens were covered and the left eyes served as the auto control eyes. The covered eyes of the control group received 20μ L 1g/L bovine scrum albumin (BSA), 10 chickens aggregated. The covered eyes of the experimental group received bFGF (1, 2, 4ng)+20μL 1g/L BSA, with each dosage on 10 chickens. The eyes in the two groups were continually covered for 1 week. The expression of VIP on retinal-choroids-sclera was investigate by streptavidin-peroxidase (SP) immunohistochemistry staining and reverse transcription-polymerase chain reaction (RT-PCR), The data were analyzed by analysis of variance (ANOVA) between the control group and experimental group and by analysis of SNK-q among each dosage in the experimental group.
RESULTS: VIP widespread expression existed on photore- ceptor-outer, inner nuclear layer, inner plexiform layer of retinal and choroids; weak expression existed on ganglion cell layer and nerve fiber layer. The expression of mRNA of VIP in auto control eyes between groups was not statistically significant (P〉0.05). In comparison with the covered eyes of control group, the expression of mRNA of VIP in experimental group were significant up-regulated (P〈0.01) , showing a dose-dependent manner (P〈0.05).
CONCLUSION: Exogenous bFGF could up-regulate the expression of VIP on retinal-choroids-sclera which may play an antagonistic action in ocular enlargement of chicken FDM.
